Mechanism: BDNF/TrkB activation via covalent attachment to surface cysteines on TrkB

extracellular domain (Cys345, Cys300 in CRD2). Reversible thioether bond prevents ligand

dissociation, extending downstream ERK1/2 and PI3K/Akt signaling for weeks.

Projected Dosing: 300 mcg SC every 14–21 days (projected); 150 mcg maintenance

Duration: 14–21 days (t½ off ~18 days; receptor occupancy 80% at day 14)

Projected Efficacy: Working memory +25–35%; neurogenesis markers ↑ 3× vs. native Semax;

sustained anxiolysis
